Histopathological and Immunological Studies on the Differences between Spontaneous Tumors and Non-Neoplastic Lesions of Laboratory Animals

Abstract
To evaluate carcinogenic or toxic effects of test substances, we have to obtain the in-house animal data of naturally-occuring tumors including non-neoplastic lesions an the basis of histopathology. Moreover, to acquired the biological raw data, we were performed the hematological, urine, clinical chemistry analysis and natural killer(NK) cell activity check in ddY and ICR mice on both sex. A total of 291 mice composed of 144 ddY mice and 147 ICR mice which were inbred in the National Institute of Safety Research were examined to observe the incidences and the histopathological characteristics of spontaneous tumors and non-neoplastic lesions of the various organ and tissue. Autopsies were carried out at 56, 80 and 92 weeks, respectively. The results were as follows ; Life span was longer in ICR mice than ddY mice. Viabillity of the ddY male and female mice at 80 weeks were 29.2% and 41.7%, respectively. But viabillity of the ICR male and female mice at 92 weeks were 50% and 41.7%, respectively, In hematological analysis, especially white blood cell differential counts have very wide-ranged values in male and female ddY mice. Percent of animals with spontaneous tumors at 92 weeks were 100%, 87.5%, 67% and 70^ in male/female ddY and IGR mice, respectively, Average frequencies of primary tumors in lymph node and lung ; 77.5% and 77.8% in ddY male; 62.5% and 50% in ddY female; 41.7% and 25% in ICR male mice, 40% and 30% in ICR female mice, respectively. Representative non-neoplastic lesions in ddY and ICR mice on both sex were pneumonitis, pyelonephritis, hydronephrosis, cysts in ovary, hypertrophy in seminal vesicles, endometrial. glands in uterus, subcapsullar spindle cell hyperplasia in adrenal glands. In natural killer cell activity analysis, ICR mice were higher than ddY female mice. Especially NK cell activity in ICR female mice were higher than male mice but not significant. NK cell activities decreased according to the age in ICR mice. However, in case of ddY female mice, the values were relatively consistent at 6, 12, 18 and 24 weeks, respectively.
